Quite a few riders I know ride with 2 inners although I've never known them to need both - you'd have to be quite unfortunate. I either use 2 and/or take a couple of sticky patches with me too. Otherwise your kit looks reasonably complete. If you can get a multitool with a chain splitter too - it might be worth it - I do and it's come in handy before

worth talking to the other riders or else everyone is carrying loads of kit, you might be able to spread it around.

co2 inflaters are good for races etc but once used they are dead and you/others might puncture again. for the L2B, a small pump is ideal. too many people also leave empty co2 containers by the trail/road side and that annoys me (though i am by no meand implying that you would do the same!)

park super patches and their tyre levers are pretty much indestructable and highly recommended.
if you are taking a camelbak or similar, a few bits of food that you like are always good, bananas, mars bars, chocoalte covered peanuts and raisins. it doesnt necessarily have to be healthy and full of carbs just stuff that you will eat and that will taste good when you need a moral booster.
If you are not used to 3+ hours in the saddle or don't cycle that much period, good shorts would be recommended. You wouldn't go wrong with some something like Assos F1 Unos,( http://www.slanecycles.com/assos-fi-uno-s5-bibshor... )they are not the cheapest short but will last you years and give you amazing comfort. I find that in many ways I can not afford to be without them.
And a rain coat that will wrap up and fit in a back pocket is a must.
