The towpath is almost certainly all three of those, but might also be the slowest. I haven't cycled in nine months.
If I was doing it on the road, I'd go through Harpsden, up the hill by the golf course, Binfield Heath, Playhatch, Sonning Bridge and down the A4. Although that's not flat, the first half is on back roads. The flattest road route would be A321 through Wargrave and then down the A4, but that would be very busy the whole way.
