The Castle of the Knights of St John in Kos Town is worth a visit, as is the Asklepion(sp?). We also took a day trip over to Nissyros island, where they take you down into the volcanic crater in the middle of the island, it was a long day (10 hours from pick-up to drop-off) but well worth the time.
I remember Kos as a near empty little island that we visited unexpectedly on a school cruise in the 1970s.
Then it was how people picture Greek Islands, quiet, peaceful, unspoilt - I understand it's a little different now.
In January we had a storm with hailstones like golf balls! Only lasted about 5 minutes, fortunately and the sun came out again!
