So I've just bought said item and I want to stick it on the boot lid of my car. I've seen cars with the same sticker used in this way before so I know it's possible.
Question is, what's the best way of transferring the sticker onto the car's bodywork?
I'd really appreciate any help you can give me!

The vinyl one?
Put the sticker on a hard surface face up. Rub the 'backing' paper with a credit card (or similar) to get the sticker to stick in all places. Peel the backing paper and the smiley will come with it - re rub if any part doesn't stick.
Place the sticker to the body, rub the backing paper to adhere to surface and peel it off to leave sticker on the paint.
Stand back, admire your work then wish you had never posted this on the internet when you realise how easy it is

Seconded, this is how i did it with my white PH Smiley, shove the backing paper on (also useful for lining up with edges, rub with finger or whatever, peel backing paper.
Oh, clean the boot first, including a good rub to get rid of Wax/polish! then if required, wax/polish over the top afterwards!
