Advice please!

Will I need to have the engine retuned if I add Hurricane, 160TB and sports exhaust to a standard Elise S1?

No.

Buy a 421 Manifold, some new cams and maybe some headwork and then you will need a new ECU

I have add the mods, but am now getting misfire (not all the time) after driving for a while on over run and hard acceleration. any ideas?

The mods you have done will not cause your car to missfire i think you may find missfire will be down to an ignition component breaking down
