Well?

IMO, the riff would have to be either Van Halen - Ain't Talkin' 'Bout Love, or Derek & The Dominos - Layla.

Opening line?

Well there's something quite electrifying about the way Rob Halford screams, unaccompanied, at the beginning of Judas Priest's All Guns Blazing:

"Twisting the strangle grip won't give no mercy,
Feeling those tendons rip torn up and mean"

Not pretty, but the song is an assault on the senses.

Recently, in terms of 'rock' music, I'd have to say the baroque but very simple intro to 'Plug In Baby' by Muse.
'Today' by the Pumpkins also has a lovely simple, but no less memorable opening riff.
