The interior of my E-Type smells like an old car - but not in a good way!

It has never been restored so the carpets.. upholstery etc are wearing 52 years of muck.

Is a detailer what I need? Or one of those re-trimmer specialists that remove the whole interior out of the car including the carpet and give it a good old clean?

Does it need to be an E-Type specialist?

Recommendations welcome.

Carpet shampoo machine, a detailer is the best place to start

A specialist classic car detailing company is probably the best way to get it assessed .
If the carpets ,sound deadening and the filling in the upholstery are suffering from long term damp , mould or rodent soiling then replacement is the recommended option .

If the floor carpets come out easily, then they can be washed relatively easily and left to dry in the sunshine.

If the car has traditional felt underfelt underneath then that is probably harbouring much of the musty smells. If its stuck down, then it could probably be wahed in situ. If its breaking up, then it may be easier to consider replacing that - either with traditional underfelt or one of the more modern options.
